چکیده

Abstract Tobacco ( Nicotiana tabacum L.) is an economic crop and a model organism for studies of plant biology genetics. As allotetraploid generated from interspecific hybridization, tobacco has massive genome (4.5 Gb). Recently, genetic map with 45,081 single nucleotide polymorphism (SNP) markers was constructed using whole-genome sequencing data population including 274 individuals. This provides basis quantitative trait locus (QTL) mapping genomic selection, which have been widely applied to other crops but not feasible in tobacco. Based on this high-density map, we identified QTLs associated important agronomic traits, chemical compounds dry leaves, hazardous substances processed cigarettes. The LOD values major were highest followed by substances. In addition the identification molecular markers, evaluated selection models found that BayesB had prediction accuracy recombinant inbred line population. Our results offer new insights into mechanism underlying such as traits quality-related tobacco, will be able support application breeding

Effects of Marker Density, Number of Quantitative Trait Loci and Heritability of Trait on Genomic Selection Accuracy

The success of genomic selection mainly depends on the extent of linkage disequilibrium (LD) between markers and quantitative trait loci (QTL), number of QTL and heritability (h2) of the traits. The extent of LD depends on the genetic structure of the population and marker density. This study was conducted to determine the effects of marker density, level of heritability, number of QTL, and to ...

High-Efficiency Agrobacterium-Mediated Transformation of Tobacco (Nicotiana tabacum)

To improve Agrobacterium-mediated transformation of tobacco, factors influencing gene delivery, including genotype of the plant, bacterial strain, and Agrobacterium transformation procedure, were tested via direct somatic embryogenesis. Leaf tissue of three different tobacco genotypes (Nicotiana tabacum L. cvs. Samsun, and Xanthi, and N. benthamiana) we...

Coproporphyrinogenase in tobacco (Nicotiana tabacum L.).

1. Coproporphyrinogenase was extracted and purified from tobacco (Nicotiana tabacum L.). Enzyme activity was mainly located in mitochondria rather than in chloroplasts. The enzyme was purified by differential centrifugation, ammonium sulphate fractionation, calcium phosphate gel adsorption and dialysis. A 69-fold final purification was obtained. 2. An apparent K(m) value of 3.6x10(-5)m was foun...

Construction of a High-Density Genetic Map and Quantitative Trait Locus Mapping in the Sea Cucumber Apostichopus japonicus

Genetic linkage maps are critical and indispensable tools in a wide range of genetic and genomic research. With the advancement of genotyping-by-sequencing (GBS) methods, the construction of a high-density and high-resolution linkage maps has become achievable in marine organisms lacking sufficient genomic resources, such as echinoderms. In this study, high-density, high-resolution genetic map ...

Genetic Study of Agronomic Traits in Tobacco (Nicotiana tabacum L.) Using Hayman’s Graphical Analysis of Diallel

For the purpose of graphical analysis and estimation of genetic parameters related to yield and its components, six tobacco genotypes was crossed in half -diallel mating design. In this study, the F2 progenies and their parents were evaluated in a randomized complete block design with three replications. The results of analysis of variance indicated a significant difference between genotypes fo...
